Research Statement

The overarching goal of the Koehler lab is to build chemical tools and methods for studying transcriptional regulation in development and disease. We pursue this goal by building general and systematic approaches for discovering small molecules that directly modulate ?undruggable? transcription factors, including oncogenic factors arising from chromosomal translocations. We also aim to discover structurally and mechanistically novel inhibitors of enzymes that catalyze post-translational modifications of transcription factors. By targeting both classes of regulators we hope to create a comprehensive chemical toolbox for studying diseases of aberrant transcription. Where applicable, we aim to develop selected probes into diagnostic tools or therapeutic leads.
